血管性痴呆小鼠海马胆碱乙酰转移酶和其mRNA变化特征及喜得镇的影响 被引量:5

摘要 目的观测喜得镇对血管性痴呆(VD)小鼠海马神经元胆碱乙酰转移酶(CHAT)及其mRNA变化的影响,以探讨两者在VD发病中的作用及喜得镇对其影响的机制。方法双侧颈总动脉线结,反复缺血-再灌注法制备模型,药物组用喜得镇溶液灌胃,利用跳台试验和水迷宫试验观测其行为学改变,采用免疫组织化学和原位杂交技术观测小鼠海马神经元CHAT及其mRNA的表达变化。结果喜得镇组小鼠学习、记忆成绩优于模型组(P<0.01),其海马CHAT及其mRNA表达也明显增高(P<0.01)。结论喜得镇改善VD小鼠学习、记忆成绩与其恢复海马低水平的CHAT及其mRNA有关。 Objective By observing characteristics of choline acetyltransferase (CHAT) and its mRNA in neuron of hippocampus in mice with vascular dementia (VD) to explore their roles in VD pathogenesis and the effect of hydergine on them. Methods The mice were subjected for ischemia-reperfusion repeatedly on bilateral common carotid arteries by knotting to establish the VD models. Hydergine was given to the mice in medication group, the behavioral abnormalities were investigated by step-down and water maze test. CHAT and its mRNA expressions in neuron of hippocampus were observed by immunohistochemistry and in situ hybridization technique.Results Learning and memory grade in hydergine group were better than those in model group (P<0.01). The changes of CHAT and its mRNA in neuron of hippocampus significantly increased in hydergine group (P<0.01). Conclusions Hydergine could improve grade of learning and memory in mice with VD, which be related to recovered CHAT and its mRNA levels.
