摘要
目的 观察微电泳P物质(SP)及其拮抗剂对间情期雌性大鼠弓状核(ARC)神经元自发放电活动的影响,进一步探讨整体情况下SP对生殖活动的调节作用及作用机制。方法 选择成年雌性大鼠,采用自制多管微电极微电泳方法研究微电泳SP及其拮抗剂对大鼠ARC神经元自发放电活动的影响。结果 微电泳SP使70 37% (38/5 4 )的受试神经元自发放电频率减慢,2 7. 78% (15 /5 4 )神经元无反应,1. 85 % (1/5 4 )神经元表现为兴奋作用。微电泳SP过程中给予其拮抗剂可逆转SP的作用。单独微电泳SP拮抗剂则使75 .4 3% (43/5 7)神经元自发放电频率加快。结论 SP对ARC神经元具有紧张性抑制作用。提示整体情况下,SP及其拮抗剂作用于下丘脑水平参与生殖轴活动的调节。
Objective To observe the direct effects of microelectrophoresis applications of substance P (SP) and SP antagonist on the spontaneous firing actives of arcuate nucleus (ARC) neuron in female dioestrum rat,and probe into the effect and mechanism of substance P on the control action of reproduction. Methods Using homemade multibarrel microelectrodes to observe the effects of SP and SP antagonist on the spontaneous firing rates of ARC neurons of female rat. Results 70.37 percent of tested neurons (38/54) were inhibited by SP administered microphoretically. 27.78 percent of them (15/54) had no response and only 1.85 percent of them (1/54) were excited. In addition, functions of SP can be reversed by SP antagonist. SP antagonist increased spontaneous firing rate in 75.43 percent(43/57) of tested neurons. Conclusions These results demonstrate clearly that SP have a tonic influence on ARC neurons. The result also give clues that SP and SP antagonist can join in the regulative action of reproduction axis in hypothalam of live rat.
