二烯丙基二硫对结肠癌SW480细胞系生长影响的研究

Effect of Diallyl Disulfide on the Colorectal Cancer Line SW480

目的 观察二烯丙基二硫(DADS)对人结肠癌SW480细胞系生长的抑制作用。方法 体外细胞培养,用MTT法、集落形成试验进行观察,体内移植瘤抑制试验采用裸鼠移植瘤模型。结果 用不同浓度DADS ( 30 μg mL ,40 μg mL ,5 0 μg mL ,60 μg mL ,70 μg mL)处理SW480细胞48小时后,测得细胞生长抑制率为2 3.8%、43.7%、49.7%、60 .9%、66.0 % ,该药的IC50 约为5 0 μg mL ;用不同浓度DADS( 2 0 μg mL,30 μg mL ,40 μg mL ,5 0 μg mL)处理SW480细胞1 0天后,集落形成抑制率为48.7%、5 6.5 %、61 .6%、76.2 % ,IC50 为2 0～30 μg mL之间;以30mg kgDADS对SW480细胞裸鼠移植瘤进行实验,测得瘤重抑制率为62 .86% ,电镜检查可见结肠癌SW480细胞有早期凋亡改变。结论 DADS对结肠癌SW480细胞系生长具有明显抑制作用。

Objective To study the inhibition effect of diallyl disulfide (DADS) on colorectal cancer cells. Methods The MTT method and cloning experiment were used to study the anticancer effect of DADS in vitro;transformated tumor models on nude mouse were took to study the anticancer effect in vivo. Results Atfer SW480 cells were treated by DADS with different concentration (30 μg/mL,40 μg/mL,50 μg/mL,60 μg/mL,70 μg/mL) for about 48 h,the inhibited growth rate of the tumor cells were 23.8%,43.7%,49.7%,60.9%,66.0% respectively.The IC 50 of DADS was about 50 μg/mL;Treated by DADS with different concentration (20 μg/mL,30 μg/mL,40 μg/mL,50 μg/mL )for 10 days,the cloning rate of SW480 was 48.7%,56.5%,61.1%,76.2% respectively.But the IC50 of DADS was between 20～30 μg/mL; Treating mice with DADS 30 mg/kg in vivo and evaluating the transformated tumors,we found the inhibition rate of transformated tumors was 62.86%.In addition,the result from electron microscope identified apoptosis in SW480 cell. Conclusion DADS has significant inhibition effect on colorectal cancer cell 1ine SW480.
