软脂酸对胰岛细胞凋亡作用的观察 被引量:2

摘要 目的 通过观察软脂酸对体外培养的原代胰岛细胞的凋亡作用,探讨游离脂肪酸在糖尿病发病中的作用。方法 体外培养Wistar大鼠胰岛细胞,用不同浓度软脂酸(分别为0 ,0 .1 2 5 ,0 .2 5 ,0 .5mmol L)共同孵育48h ,用Tunel法计算胰岛细胞凋亡率。AO EB染色观察细胞形态变化。结果 0 .1 2 5 ,0 .2 5 ,0 .5mmol L的软脂酸均有抑制高糖刺激下胰岛细胞的胰岛素分泌,促进胰岛细胞凋亡的作用。结论 软脂酸可抑制胰岛细胞的分泌功能,促进胰岛细胞凋亡,其凋亡程度与软脂酸的浓度有关。 Objective To investigate the effect of a chronic exposure to high level palmitic acid on islet cell apoptosis, thereby to understand the role of palmitic acid in the pathogenesis of type 2 diabetes mellitus. Methods Wistar rat pancreatic islet cells were isolated and cultured in monolayer in vitro with or without palmitic acid (0.125,0.25 ,0.5 mmol/L),and the insulin concentrations in culture medium were determined by radio immunological methods. The percentage of apoptotic cells was assessed by Tunel.Morphological observation with a fluorescence microscope was conducted after double staining of the cells with AO/EB. Result Palmitic acid with different concentrations(0.125,0.25 ,0.5 mmol/L )all have the effects of inhibiting insulin secretion of pancreatic islet cells and inducing islet cells apoptosis. Conclusion Palmitic acid is capable of inducing islet cells apoptosis in dose-dependent manner
