摘要
利用国产液电碎石机(ESWL-Ⅲ/2M型)产生的高能冲击波和10-羟基喜树碱单纯或联合处理人膀胱癌细胞系(BT5637)。实验分对照组,HESW组,药物组和联合组。冲击波条件:200SW(10kV,60次/min),药物浓度(0.1μg/ml),水温37℃。采用α-32P-dCTP标记C-myc癌基因探针,通过Northern印迹杂交技术,分析四组癌细胞C-myc癌基因表达。结果表明:对照组C-myc癌基因表达明显,其转录产物为2.7kb,而实验各组C-myc癌基因表达完全被抑制。结论:HESW,10-羟基喜树碱及其联合对人膀胱癌细胞的生长抑制作用与C-myc癌基因表达有密切关系。
Human bladder tumor cell line (BT5637) was managed by HESW(high energy shock waves,electrohydrulically generated, ESSL-Ⅲ/2M type) alone or in combination with 10-hydroxy camptothecimun(an anti-tumor drug).The experiment includes a comtrol,a single HESW,a singal drug and a combination group.200sw(10kV, 60time/min)and 0.1ug/ml 10-hydroxy camptothecinum were given to the HESW, drug and combination group respectively. The expression of C-myc oncogene in these groups was analysed by northern blot hybridezation with a- ̄(32)p-dCTP labeled C-myc gene probe. The Result showed that C-myc oncogene expression in the cotrol group has been very obvious,Its transcription product being 2.7kb. But the C-myc oncogene expression in the experimental groups were inhibited completely.
