摘要
目的 探讨 P1 6 基因在腮腺多形性腺瘤发生、发展及在判断预后中的作用。方法 采用 SABC法行免疫组织化学染色 ,利用计算机图像处理系统 ,对 P1 6在腮腺多形性腺瘤中的蛋白表达进行定量检测 ,结合肿瘤细胞核形态学参数等相关数据 ,进行统计学分析处理。结果 多形性腺瘤的发生与 P1 6抑制基因密切相关 ,P1 6蛋白表达与肿瘤细胞分化程度紧密相关 ,与肿瘤生长时间呈负相关 ,P1 6蛋白的低表达、缺失可导致肿瘤的恶性发展 ,使其生物学行为恶化。结论 P1 6 抑制基因与多形性腺瘤的生物学行为关系密切 ,P1 6 蛋白可作为肿瘤分级和区别肿瘤增殖潜能的有效指标。 P1 6 蛋白缺失及低表达者应视为预后较差 。
Objective To investigate the roles of P 16 gene in the development and progression of pleomorphic adenoma.Methods Computer image analysis techniques was used to quantitate the result of immunohistochemical reaction of P 16 protein and nuclear morphologic features of neoplastic cells, the expression of P 16 gene was detected by SABC immunohistochemical staining technique.Results P 16 expression is closely related to the degree of differentiation of neoplastic cells in pleomorphic adenoma.Accumulation of p16 is negatively correlated with tumor growth time. Low expression or/and deletion of P 16 protein greatly enhance malignant progression of disease, make the biological behavior more malignant with a poor prognosis.Conclusion The biological behaviors of pleomorphic adenoma are closely correlated with P 16 expression, immunohistochemical detection of P 16 gene expression in tumor tissues may be used as a method to judge the multiplication potentiality of tumor cells. Low accumulation of P 16 protein is at high risk of developing carcinoma,closely observation should be performed in these patients after surgical treatment.
