摘要
通过设计正交试验,对重组Pichiapastrois酵母利用YEPD培养基表达纳豆激酶基因的发酵条件进行了研究,确定最适条件:pH值6·0,培养温度30℃,培养时间36h,诱导时间84h,甲醇添加量1·0%,其中培养温度为主要影响因素。在该条件下进行重组子的培养及诱导表达,SDS-PAGE电泳结果表明外源蛋白的表达量占菌体蛋白的10%。
In this study, fermentation conditions for yeast Pichia pastrois containing nattokinase gene were investigated through orthogonal test using YEPD culture medium. The results showed that the expression protein was about 10% of total protein when cultured at pH 6.0, 30℃ for 36h and induced with 1% of methanol for 84h. It is found that the culture temperature was the most significant among all the factors.
