摘要 介绍了3种测定克拉维酸含量的方法,即高效液相色谱法、紫外分光光度法与生物效价法,3种方法测定结果进行分析研究,得出三种方法之间具有较好的相关性。高效液相色谱法及紫外分光光度法均基于克拉维酸与咪唑反应衍生后的衍生物在312nm处有特征吸收,且衍生后的色谱图在反相高效液相色谱柱上可有效避免发酵液中其他组分的干扰;生物效价法采用KlebsiellapneumoniaeATCC29665作为指示菌。由于3种方法测定结果之间具有相关性,可以根据不同的实验条件及实验目的选用不同的检测方法。 High performance liquid chromatography (HPLC), spectrophotometry and bioassay methods were used to determine the concentration of clavulanic acid in fermentation broth in the present paper. The relationship between the results of these three methods was investigated. It is found that the results of the three methods are correlation. HPLC and spectrophotometry methods were based on the absorbance at 312 nm from the reaction of clavulanic acid imidazole. This derivative chromatographs from reverse-phase HPLC columns clear of interfering components in fermentation broth while the bioassay method use Klebsiella pneumoniae ATCC29665 as the indicator organism.Because of the correlation of the results determined by three methods we can select different methods based on different conditions and purpose.
