摘要
将胎龄为18~32周的水囊引产死胎之脑垂体进行体外培养,然后分为培养细胞移植组、未培养垂体组织块移植组和培养液对照组,将移植物移植于BALB/c裸鼠的背侧颈部皮下。定期取血清用酶联免疫吸附法(ELISA法)测卵泡刺激素和黄体生成素。三组间激素水平的差异均有显著性,培养细胞移植组30天内激素水平一直维持于较高水平。表明经培养的垂体细胞行动物背侧颈部皮下移植,能在动物体内存活,并具分泌功能。
To find a simple and effective treatment for Sheehan's Syndrome, we investigated the transplantation of human fetal pituitary in nude mice. Six nude mice were divided into three groups: transplanted with cultured pituitary cells, with pituitary tissue fragments, and with only RPMI 1640 medium. Blood samples were collected periodically and the FSH and LH levels were detected by ELISA method. The differences between all 3 groups were significant. The hormone levels in the cultured pituitary cell group were persistently high in 30 days. The results showed that the cultured pituitary cells could survive in vivo and could secrete hormones when the grafts were transplanted subcutaneously in the neck of the nude mice.
