bFGF、αⅤβ3和NFκB在hOEC中的表达及其与血管生成关系的研究 被引量:6

摘要 目的:研究碱性成纤维生长因子(bFGF)、整合素αⅤβ3和核因子κB(NFκB)在人卵巢上皮性癌(OEC)组织中的表达及其与血管生成的关系·方法:原发性人卵巢上皮癌(hOEC)36份、卵巢良性肿瘤及正常卵巢15份,应用免疫组化方法(ABC法)检测bFGF、整合素(integrin)αⅤβ3、NFκB、微血管密度(microvesseldensity,MVD)在OEC、卵巢良性肿瘤及正常卵巢组织中的表达及其与临床病理特征的关系.结果:bFGF、αⅤβ3、NFκB和MVD在各种组织中的表达,36份OEC中的阳性表达率分别为66·67%、66·67%、72·22%和(43·98±33·73),在良性肿瘤组织中的表达率分别20·00%、6·67%、26·67%和(13·08±9·07);在正常卵巢组织的表达率分别为0·00%、0·00%、13·33%和(7·10±5·42),OEC组各指标的阳性率均显著高于良性卵巢肿瘤组及正常对照组(P<0·05).OEC中bFGF、αⅤβ3、NFκB、MVD随着肿瘤分化程度的降低、FIGO分期的增加、肿瘤转移范围扩大,各指标的表达均逐步增高(P<0·05).OEC中bFGF、αⅤβ3、NFκB与MVD的表达的相关系数分别为0·69、0·67、0·62、各指标与MVD均显著相关(P<0·01).NFκB阳性表达者bFGF、αⅤβ3的阳性表达率分别为92·86%和85·71%,显著高于NFκB阴性表达者bFGF、αⅤβ325%的表达率(P<0·05).结论:bFGF,αⅤβ3,NFκB在OEC中的表达显著增高,与OEC血管生成密切相关.bFGF、αⅤβ3在OEC中的表达与NFκB表达呈正相关,NFκB可能通过对bFGF、αⅤβ3的正向调节作用,进而影响OEC血管生成. Objective: To study the expression of basic fibroblast growth factor(bFGF), integrin (αⅤβ3) and nuclear factorκB(NF κB) in human ovarian epithelial carcinoma(OEC) tissues and their relationships with angiogenesis.Methods: 36 samples of OEC tissues and 15 samples of ovarian benign tissues and normal ovary tissues were selected.The expression of bFGF, αⅤβ3, NFκB and MVD in OEC, ovarian benign tumors and normal ovaries were detected by immunohistochemical staining (ABC method), and their relationship with the clinical pathological characters were studied. Results: The expression rates of bFGF, αⅤβ3, NFκB and MVD in OEC were higher than those in ovarian benign tumor and normal ovarian tissues.The expressions of bFGF, αⅤβ3, NFκB and MVD in OEC increased with the decrease of differential grade, the increase of FIGO stage and extension of metastasis.There were significant relationship between bFGF, αⅤβ3, NFκB and MVD(P<0.05).The expressions of bFGF and αⅤβ3 in NFκB positive group were 92.86% and 85.71% respectively, higher than those in NFκB-negative group. Conclusions: There are high expressions of bFGF, αⅤβ3, NFκB and MVD in OEC which are significantly related to angiogenesis, and related to advance of diseases and prognosis of patient with OEC.The expressions of bFGF, αⅤβ3 are Positively related to NFκB, NFκB which may upregulate the expressions of bFGF, αⅤβ3.Thus these factors may enhance angiogenesis of OEC.
