摘要
目的探讨肺间质成纤维细胞基质金属蛋白酶2(MMP-2)在肺纤维化中的表达及意义。方法清洁级Wistar大鼠50只,随机分为对照组和实验组,实验组气管内注射SiO2悬液,对照组代以等剂量生理盐水。注射后1,3,7,14,28d分离和提取肺间质成纤维细胞,应用免疫细胞化学和逆转录多聚酶链式反应(RT—PCR)方法,观察其MMP-2蛋白及其mRNA的动态变化。结果①SiO2作用后3d成纤维细胞MMP-2表达增高,达对照组的2.423倍(t=5.205,P<0.01),此后一直维持高水平;②MMP-2mRNA的转录从SiO2作用后1d就开始升高。结论在肺纤维化早中期,肺间质成纤维细胞高表达MMP-2。
Objective To study the expression and its significance of MMP - 2 on fibroblasts of pulmonary fibrosis model rats. Methods 50 pathogen- free Wistar rats were divided randomly into experiment group and control group. Rats in the experiment group were instillated with SiO2, while control group with normal asline instead. Pulmonary interstitial fibroblasts were isolated at 1,3,7,14,28th day after SiO2 exposure. The protein and mRNA expression of MMP- 2 were evaluated by Immunocytochemistry and RT- PCR. Results ①The MMP-2 protein expression of fibroblasts increased 2.423 times (t = 5.205,P<0.01) higher than that of control in 3 day, and then remained at the high level. ②The mRNA expression of MMP - 2 was enhanced in fibroblasts in 24 hours after SiO2 instillation. Conclusion mRNA expression of MMP - 2 in fibroblasts are up-regulated in fibrosis silicotic model rats.
