鸡类囊胚获得及其生物学特性研究 被引量:4

摘要 鸡胚是研究胚胎发育、组织分化、基因表达与调控良好的实验模型, 是动物功能基因组学研究的重要材料. 实验选取白来航蛋鸡所产新鲜种蛋, 采用纸环法收集胚盘, 机械打碎后进行悬浮培养, 获得了哺乳动物囊胚样结构, 命名为类囊胚. 体外培养12~24 h是类囊胚发育的高峰期, 其直径显著增加近1倍. FBS可促进类囊胚的发育, 而卵黄在12 h前对类囊胚发育没有明显影响, 但到24 h时可促进类囊胚的贴壁生长. 经碱性磷酸酶(AKP)染色初步显示类囊胚中含有大量碱性磷酸酶活性的胚胎干细胞, 胚胎干细胞特异性表面标志SSEA-1免疫荧光染色后呈阳性, 进一步肯定了类囊胚中ES细胞的存在, 最终从贴壁类囊胚中分化出神经样和成纤维样等多种类型细胞. 由此可以得出: 悬浮培养鸡胚盘细胞可以形成含有胚胎干细胞的、类似哺乳动物囊胚样的结构, 并能保持其内胚胎干细胞的特性和分化潜能.
