抗雌二醇单克隆抗体的制备与鉴定 被引量:3

摘要 活化雌二醇(E2)C3处的酚-OH,与牛血清白蛋白(BSA)偶联,制备得E2的完全抗原,利用完全抗原免疫动物,采用单克隆抗体技术,经过细胞融合、克隆筛选、扩大培养、动物体内诱生腹水等过程,最后获得了E2的单克隆抗体,对纯化的单克隆抗体的性质鉴定,结果显示:其抗体效价为1:106,抗体属于IgG2a型;分子量为179000Da;E2单克隆抗体的亲和常数K2.9108L/mol。 Estradiol (E2) was bound to bovine serum albumin (BSA) to form a complete antigen (E2-BSA), after the hydroxy (-OH) at the position of C3 was activated. A BALB/c mouse was immunized with the mixture of E2-BSA and Freund adjuvant. Monoclonal antibody-producing hybridoma anti-E2 was prepared by fusion of SP2/0 murine myeloma cells with spleen cell isolated from the immunized mouse. A monoclonal antibody (McAb) specific for E2 was produced in vivo by the hybridoma anti-E2. Analysis revealed that anti-E2 McAb (E2-McAb) was of the IgG2a type, with the molecular weight179000 daltons. The titer of E2-McAb was 1:106, and the affinity constant of constant of E2-McAb was 2.9×108L/mol.
