摘要
采用超声、差速离心法制备肾综合征出血热病毒(HFRSV)抗原。通过与免疫荧光法(IFA)和捕获IgMELISA法(MacELISA)比较,分析了病人临床四个期血清抗体阳性率,对3组不同地区的正常人群HFRSV抗体阳性血清作抑制试验,以及比较IFA和ELISA血清抗体几何平均滴度证实本方法是特异和灵敏的。用这一方法对野鼠型和家鼠型HFRS病人血清,与两型病毒作交叉ELISA试验,结果呈单向反应;采用胞膜荧光反应为主的感染细胞制备抗原,可提高其分型效果。
The Indirect IgG ELISA was improved by using a rdfined hemorrhagic fever with renal syndrome virus(HFRSV) antigen by Supersonication of the infected cell culture and purified by differential speed centrifugation, and by using and optimalized quantity Protein content of the antigen. The improved method gave higher antibody titres. Its specificity and sensitivity were demonStrated by its comparison with IFA and the IgM-capture ELISA of a panel of positive patient sera collected at each of the four Clinical phases, respectively, and by the Inhibition ELISA test of the positive human sera collected from three healthy populaTions, and by comparative studies of IFA titres with consecutive serial sera from designated patients. PreliMinary results showed that there was a one-way reaction in cross-ELISA analysis of sera form patients with the Hantaan-type Infection and a bilateral cross reaction with the Seoul-type infection. It was found that using antigen prepared from infected cells with a predominatnt immunofluorescent surface reaction, a more clear-cut serotyping result could be obtained.( Institute of Virology, Chinese Academy of Preventive Medicine, Beijing100052)( Anhui Province Feng Tai County People′s Hospital)
