In vitro osteogensis and calcification of rat calvaria osteoblasts by ALPase elecfronmicroscopic cytochemistry

摘要 应用碱性磷酸酶免疫组织化学和电镜细胞化学实验方法,观察体外培养鼠成骨细胞骨生成过程中基质小泡的形成,分泌及诱导基质钙化的细胞学变化。证实成骨细胞在体外适宜环境下,同样具备骨生成的能力,其成骨过程与体内的膜内成骨过程相似。 Abstract The procedure of in vitro osteogenesisand calcification of rat calvaria osteoblasts wasstudied by the methods of ALPase immunohis-tochemistry and electron microscopic cyto-chemistry.The formation,secretion and ma-trix mineralization functions of matrix vesicleswere observed.It was concluded that os-teoblasts have the capacity to differentiate intoosteocyte and formed calcified bone tissue in a suitable enviroment in vitro.The calcificationprocedure is similar to that of intramembra- nous ossification in vivo. For clinical rele-vance,the results will play an important rolein the treatment of osteoporosis and long dis-tance bone defects.
