脑缺血及再灌流时脑组织自由基水平的研究 被引量:13

Experimental study on level of free radi-cal In brain ischemia-reperfusion

摘要 本实验选用60只雄住大鼠,体重250g~40g,随机分为6组:①对照组;②缺血3Omin组;③缺血3Omin再灌流30min组;④缺血30min再灌流6Omin组;⑤缺血3Omin再灌流12Omin组;⑥治疗组。每组各10只。通过制备大鼠完全脑缺血模型,测定各组脑组织中丙二醛及超级化物歧化酶的含量,并结合电镜检查。结果显示,缺血组及再灌流各组脑组织中丙二醛含量均高于对照组(P<0.05);而超氧化物歧化酶含量均低于对照组(P<0.01),同时电镜观察显示脑组织水肿及退行性改变。应用“仙台合剂”治疗则能明显减轻上述改变,对缺血及再灌流后引起的脑组织损伤有保护作用。 Sixty male SD rats were equally devidedinto six groups. An animal model with brainischemia- reperfusion had been established。Malonyldidehydf(MDA)and superoxide dis- mutase(SOD)were mesured in brain ho-mogenate,and at the same time,ultrastructureof brain tissue was observed. The result showthat MDA in ischemia and ischemia-reperfu- sion groups increased significatly over that ofexperimental control group(P>0.05),SODin ischemia and ischemia- reperfusion groupsdecreased significantly that of experimentalcontrol group(P>0. 01)and the injuredchanges existed pathologically。MDA in treat-ed group was obviously decreased comparedwith of ischemia 30’- reperfusion 60'group(P>0.01),SOD in treated group was obviouslyincreased compared with that of ischemia 30'- reperfusion 60'group(P>0.0l)and thepathology was ameliorated.
