摘要
目的观察慢性重型肝炎及肝衰竭患者血浆对C3A细胞株的影响,为研究应用C3A细胞株的生物反应器进行生物人工肝支持治疗提供实验依据。方法用100%正常人血浆(normalhumanplasma,NHP)和100%慢性重型肝炎患者的血浆(chronicseverehepatitisplasma,CSHP)培养C3A细胞,四甲基偶氮唑盐法(Methylthiazolyltetrazolium,MTT)检测细胞活力,通过3H-亮氨酸掺入法检测细胞蛋白合成速率,Western-blot分析比较白蛋白合成功能,从细胞活力和蛋白合成方面研究慢性重型肝炎患者血浆对C3A细胞的影响。结果①连续5天检测100%CSHP培养的C3A细胞活力均明显高于100%NHP组,(t值分别为6.26、4.89、3.55、3.94、6.17,P值均小于0.01);②第48h100%CSHP培养的C3A细胞蛋白合成速率高于100%NHP组,结果差异有显著性(t=5.93,P<0.01);③培养24h后Western-Blot分析可见100%CSHP组白蛋白表达量明显高于100%NHP组。结论CSHP对C3A细胞的增殖和蛋白合成功能有促进作用。
Objective To investigate the effects of plasma from patients with chronic severe hepatitis on C3A cell line in vitro, which can provide experimental data for C3A cell derived bioartificial liver. Methods C3A cells were cultured in normal human plasma(NHP) and chronic severe hepatitis plasma(CSHP). We detected the cell activity by MTT, protein synthetic rate by 3H-leucine incorporation, albumin synthesis by western blot. Results ①The activities of C3A cells cultured in CSHP were higher than that in NHP for successive 5 days (P<0.01); ② The protein synthetic rates of C3A cells in CSHP for 48h were higher than that in NHP(P<0.01); ③ The expressions of albumin by Western-blot in C3A cells cultured in CSHP for 24h were much more than that in NHP. Conclusion CSHP can promote the proliferation and synthesis of C3A cell.
