摘要
作者测定了各组动物肝缺血再灌注前后肝组织及血中丙二醛(MDA),谷胱甘肽过氧化物酶(GSH-px)及超氧化物歧化酶(SOD)的变化,并行组织学、硝酸镧示踪和超微结构观察。结果表明:肝缺血再灌注后有氧自由基的产生及抗氧化酶的抑制,并发生缺血再灌注损伤。白细胞浸润、氧自由基的产生及肝窦内皮细胞,Kupffer细胞的损伤可能在这一过程中起重要作用。MP可减轻这种损伤。
This study investigated the effects of pre-treatment
with methylprednisolone(MP)onliver tissue injury associated with hepatic
is-chemia-reperfusion. Twenty-nine rabbits wererandomized into three groups. Group A
re-ceived saline as a control.Group B was pro-cessed in a sham operation. Group C was
pre-treated with MP(30~40mg/Kg).After onehour of hepatic reperfusion which followedForty-five
minutes of complete ischemia,a sig-nificant increase in the level of liver tissueMDA(P<0.01),and
a decrease in tissueGSH-Px(P<0.01)and blood SOD activities (P<0.01)occured .After pretreating
the rab-bits With MP,the devation of MDA(P<0.01)was suppressed .during the period of
reperfusion following ischemia. Histopatholo-gical and ultrastructural examinations showedan
infiltration of a great number of neu-trophilic leukocytes,increased permeability ofcell
membrane,and damage of liver cells,espe-cially that of epithelial and Kupffer cells.MPcould
significantly lessened the above tissueinjuries.These findings suggest that
hepaticischemia-reperfusion cause liver tissue injury.Infiltration of leukocytes,oxygen-free
radi-cals,and,damage of epithelial and Kupffercells may play an important role in hepatic
is-chemic-reperfusion injury. Pretreatment withMP has a protective effect on this type of tis-sue
injury.
