应用流式细胞仪测定骨巨细胞瘤的DNA含量 被引量:2

摘要 用流式细胞仪对44例骨巨细胞瘤(GCT)DNA含量进行了分析,并对其中2例GCT进行GCT肿瘤相关抗原和DNA含量双参数测定。结果42例Ⅰ、Ⅱ级GCT中有16例异倍体,1例肺转移和2例Ⅲ级GCT为异倍体。统计学分析表明:(1)GCTⅠ级与Ⅱ级间的增殖速度与异倍体比例均无显著性差异。(2)Ⅰ、Ⅱ级原发和术后复发GCT之间无论是异倍体所占比例和增殖状况都有显著性差异。(3)Ⅰ、Ⅱ级GCT经体外培养后细胞增殖加快,具有GCT肿瘤细胞相关抗原的细胞比例增加。提示GCTⅠ、Ⅱ级间形态学上的差异还不能完全反映其内在本质;证明流式细胞术在临床上可对GCT局部侵袭性评价提供客观线索和依据;支持GCT中存在肿瘤细胞和非中瘤细胞成分,只有肿瘤细胞成份在体外培养中增殖存活。 AbstractThe DNA content of 44 giant cell tumors (GCTs)was determined by flow cytometry and simultaneousannalysis of cell surface antigens and cellular DNA wasperformed in two of these GCTs. Two grade Ⅲ GCTs,one with lung metastases, were all aneuploid. Out of42 grades Ⅰ and Ⅱ GCTs,16 were aneuploid, statisti-cally, there was no difference in proliferative activityand rate of aneuploidy between grade Ⅰ and grade ⅡGCTs, but significant difference between primary andlocal recurrent GCTs existed. After 5 passages in cul-ture the proliferative rate of cells in 13 GCTs increasedsignificantly, the rate of cells bearing GCT associatedantigen stepped up and in 2 GCTs tested specially. Allthese findings demonstrate : (1) the morphological dif-ference of these tumors was not remarkable (2) DNAdeterminations as an adjunct to conventionalhistopathologic assessment may provide objective clini-cally relevant information for prognosis, (3) tumor andnon-tumor cells co-exist in GCTs and only tumor cellscould survive in long-term culture in vitro.
