摘要
为探讨更年春方(GNC)预防实验性原发性骨质疏松的作用机理,采用10~12月龄SD雌性大鼠去卵巢建立骨质疏松模型,用GNC灌药4个月,RT-PCR法检测胫骨骨骺ERα、ERβ、OPG、RANKL、osf2/cbfal与TRAP mRNA的表达.结果:大鼠去卵巢后,血清E2水平及胫骨骨骺ERα、ERβ、OPG及osf2/cbfal mRNA的表达均明显下降,RANKL与TRAPmRNA的表达明显增高.GNC治疗后,血清E2水平无明显变化,胫骨骨骺ERβ、OPG及osf2/cbfal mRNA的表达均明显增高,RANKL与TRAP mRNA的表达明显下降.提示GNC可能通过上调胫骨骨骺ERβ mRNA的表达,有促进成骨细胞、抑制破骨细胞的作用,从而对实验性原发性骨质疏松有预防功效.
To study the mechanism of “Gengnianchun Liquid” in the prevention of experimental primary osteoporosis, SD female rats were oophorotomized to make osteoporosis. After treatment by “Gengnianchun Liquid” for 4 weeks, ERα, ERβ, OPG, RANKL, osf2/cbfal and TRAP mRNA expression were detected. Results: In oophorotomized rats, E 2 , ERα, ERβ, OPG and osf2/cbfal mRNA reduced, while RANKL and TRAP mRNA expression increased. After treatment by“Gengnianchun Liquid”, E 2 changed little, ERβ, OPG and osf2/cbfal mRNA expression up regulated, RANKL and TRAP mRNA expression down regulated. It is indicated that “Gengnianchun Liquid” may up regulate ERβ mRNA expression to prevent osteoporosis.
