冷藏对离体缺血组织的保护作用组织化学观察 被引量:36

PROTECTIVE EFFECT OF COOLING IN DE-TACHED ISCHEMIC TISSUE:A HISTOCHE-MICAL EVALUATION

摘要 组织离体后缺血、缺氧,细胞逐步变性,最后步入不可逆性变性而导致坏死。冷藏可延缓新陈代谢过程,推迟不可逆变性到来,从而较久地保存组织活力已被证实而应用。对于断肢、断指、皮瓣或其他移植物放置4℃容器内以保存组织活力是通行的方法。为了观察其有效程度、探索合理的再植或移植的时限,本研究以组织化学手段观察离体断指不同组织中的ALP,ATPase及SDH的变化,从功能角度判断冷藏保护的有效程度和有效时限。结果显示肌肉与血管壁细胞对缺氧最为敏感。经过冷藏的肌肉与断指,合理缺血时限分别为2与7天,可能争取的缺血时限分别为11与12天。 AbstractDetached tissues will undergo degenerativeprocess and lead to necrosis finally due to lackof blood and oxygen supply,It has been provedthat ccoling may lower metabolic processand protect the tissues from necrosis in a limi-ted time.Storting the amputated digit or limb,composite tissue flap or transplants in a coolcontainer to prelong the ischemic time is a pro-ven measure used effectively. This experimentis designed to detect the effectiveness in delay-ing the degenerative process and to assess thereasonable ischemic time limit by histochemis-try method.The amputat ed digits were dividedinto two groups and were kept in 15℃ incubatorrefrigerator separately.Specimens were takenfrom each group every 6 hours.The ALP,ATPaseand SDH of the specimens were stained andtheir density of staining observed to assess thevitality of the tissues on a functional base. Bycomparison of the vessel wall is most sensitiveto ischemia(anoxia)than the skin and its ap-pendages. The reasonable ischemic time for coolpreserved muscle and amputated digit are 2and 7 days respectively while the ischemic timeredering low level tissue vitality are 11 and12 days respectively.
