摘要
目的探讨大黄对急性胰腺炎大鼠肠黏膜屏障功能的保护作用。方法 SD 大鼠32只,采用3.5 mg/L 牛磺胆酸钠逆行胰胆管注射建立急性胰腺炎模型后,随机分为对照组(n=16)及治疗组(n=16)。对照组给予全肠外营养支持,治疗组给予相同配方的全肠外营养支持,并予大黄灌胃。于治疗后第1及第5d 处死大鼠,检测肠道通透性、吸收功能、空肠黏膜蛋白、DNA 含量以及肠道细菌易位率。结果治疗后第1、5d,治疗组肠道通透性均显著低于对照组(P<0.05;P<0.01);治疗后第5d 肠道吸收功能显著高于对照组(P<0.05),空肠黏膜蛋白及 DNA 含量显著高于对照组(P<0.01)。肝脏、脾脏及血中细菌培养阳性率显著低于对照组(P<0.05)。结论大黄对急性胰腺炎大鼠肠黏膜屏障具有保护作用。
Objective To study the protective effect of rhubarb on intestinal barrier function in rats with acute pancreatitis.Methods 32 male SD rats received injection of sodium taurocholate solution(3.5mg/L) into the pancreatic duct and were randomly divided into control group(n=16) and treatment group (n=16).Animals in control group received total parenteral nutrition (TPN),animals in treat- ment group with the same TPN formula as control group and rhubarb was given orally at a dose of50 mg/kg.Rats were sacrificed on d1 and d5 of TPN.Concentration of xylose and fluorescein isothiocyanate (FITC) dextran in superior mesenteric vein(SMV),protein and DNA con- tent in jejunal mucosa were determined.Samples from SMV,mesenteric lymph nodes,pancreas,liver,spleen were harvested for cul- tures.Results FITC-dextran concentration in treatment group was significantly lower than control group on d1 and d5 (P<0.05 or P< 0.01 respectively),but on d5 xylose concentration in treatment group was significantly higher than control group(P<0.05).Protein and DNA content in jejunal mucosa in treatment group were significatly higher than control group on dS( P<0.01).Positive cultures in liver, spleen and blood in treatment group were significantly lower than control group(P<0.05).Conclusion Rhubarb might prevent the en- hancement of intestinal permeability and bacterial translocation,but improved intestinal absorption function during TPN in rats with acute pancreatitis.
