摘要
TNFα系单核-巨噬细胞产生的细胞因子,具有明确促进纤维母细胞增殖的作用,但对胶原产生的影响文献报道不尽相同。本文研究并比较了重组TNFα对NIH/3T3细胞及大鼠肝贮脂细胞增殖,Ⅰ、Ⅲ型前胶原基因表达的影响,结果发现TNFα具有促进上述两种细胞增殖作用,且具剂量依赖性。原位杂交显示,在单个细胞水平,TNFα可抑制或降低Ⅰ、Ⅲ型前胶原mRNA水平的表达。本研究提示肝贮脂细胞与纤维母细胞类似,接受TNFα调控,TNFα在肝纤维化始动激活及调控中具重要作用。

on collagen production was controversial.Theeffect of recombinant TNFα on NIH/3T3 cell and
rat lipocytes proliferation as well as typeⅠand type Ⅲ procollagen gene expression were
studied.The results showed that TNFαstimulated the proliferation of NIH/3T3 cell and lipocyte
in dose-dependent and inhibited thetypeⅠand type Ⅲ procollagen expression at the level of
individual cell. The study revealedthat rat liver lipocyte is comparable with NIH/3T3 cell in the
presence of rTNFα. TNFα isone of the important mediators in the fibrosis initiation and
regulation.
