人表皮细胞的体外改良培养

An improved method to culture epidermal cells

目的:探讨人工纯化法去除成纤维细胞对人表皮细胞生长的影响。方法:原代培养人表皮细胞第5天时,采用人工纯化法即用0.25%胰蛋白酶+0.04%EDTA(1∶1)消化细胞2～10min,除去成纤维细胞后,观察表皮细胞生长情况并用台盼蓝染色检测其存活率。结果:细胞存活率为87%,存活率较高。生长状态较传统方法培养的表皮细胞生长状态好。结论:人工纯化法除去成纤维细胞为原代培养表皮细胞的方法改良,有利于表皮细胞更好的生长。

Objective: To investigate an improved method to culture epidermal cells.Methods: To remove fibroblasts,epidermal cells were manually purified by 0.25% trypsin plus 0.04% EDTA(1∶1).The growing state of epidermal cells was observed and the survival rate of epidermal cells was detected by tryphan blue staining.Results: The survival rate of epidermal cells was 87%.The growing state of epidermal cells cultured after manually purification was better than that of epidermal cells cultured by traditional methods.Conclusion: Manually purification to remove fibroblasts is an improved method to culture epidermal cells primarily.
