摘要
肿瘤坏死因子(TNF)是由单核-巨噬细胞产生的单核细胞刺激因子,最近有人报道T及B淋已细胞也能产生TNF,提示TNF-α来源的多样化。本研究提示用TPA、OK432刺激诱导只有单核细胞产生TNF-α,而PHA-P可使淋巴细胞和单核细胞均产生TNF-α。用OK432对不同类型白血病细胞进行诱导,发现M_4及M_(5b)型白血病细胞可产生大量TNF-α。细胞表面CD_(14)抗体阳性率越高TNF-α产率也越高。提示TNF-α的产生可作为单核细胞分化的一种指标,用以对细胞因子与白血病增殖作深入的研究。
he in vitro production of tumor necrosis factor-α(TNF-α) induced by the streptococal preparation OK432was examined to assess the monokine production capaci-ty of the leukernic cells. The TNF-α-producing cells innormal peripheral blood were monocytes when stimulat -ed with OK433 or 12-0-tetradecanoyl-phobol 13-acetate(TAP) , while both lymphocytes and monocytes could be stimulatcd to produce TNF-α with PHA-P.The capacity of the leukemic cells from 43 patientswith acute lymphoblastic leukemia ( ALL) , 13with acute nonlymphocytic leukemia (ANLL) and 2with chronic myelogenous leukemia (CML) to produceTVFα was investigated in the present study. In 43 ALLpatients. only 7 could produce relatively small amount ofTVFα. Among them, 4 were common ALL, 2 undiffer-enuated ALL and 1 B-cell ALL, two of them also ex-pressed myeloid antigens on the same leukemic cells, 2cases with undifferentiated type of ALL under one yearof age. In 18 cases of ANLL, 2 of 3 M_1 and 1 of 3 M_2could produce TVFα, all cases with M_4 ( 6 cases) andM_(5b) (2 cases) subtype revealed large amount of TNFαproduction, while none of M_(5a) (2 cases) secreted TNFαin vitro. Both of CML patients showed srnall arnount ofTNFα production. The capacity to produce TNFα inleukemia cells was wcll correiated with the exnressioii ofmonocyte specific CD14 antigen. so the TVFa produc-tion in childhood leukemia would be one of the cbarac-teristic of leukemic cells with monocytoid differentiation.
