摘要
介绍高效液相层析(HPLC)技术微量和快速检测珠蛋白体外生物合成的新方法,并运用此方法对各种类型的地中海贫血患者的网织红细胞中各种珠蛋白链的合成情况进行了研究,结果表明该方法灵敏、省时,重复性好,能可靠地检测出细微的生物合成差异,并首次在蛋白水平上证实中国人中常见的一种β地中海贫血基因βIVSIInt654C→T导致的是β ̄+-地中海贫血。
his paper describes a new technique of globin chainbiosynthesis, i, e, microglobin chain biosynthesis byrapid HPLC procedure for the studies on globin chainbiosynthesis in reticulocytes of a number of cases of vari-ous types thalassemia syndrome. The data showed thatthis technic had the advantages of sensitivity and time-saving. Particularly, it proved to be an effective rnethodto detect the small difference of globin chain syntheticratio with reliability and reproducibility. This paper alsoreports the result uf globin protein syntheais in the pa-tient with β-thalasseinia gene IVs-2-nt. 654 C→T,thatcommon Chinese mutant leads to a β-thalassemia at theprotein level.
