摘要
目的:研究蛋白激酶C(PKC)在NIH3T3细胞中,对G2/M期的关键调节因子促成熟因子(MPF)的调控作用,检测MPF两种亚基p34cdc2和cyclinB在翻译水平的改变。方法:用aphidicolin,流式细胞仪检测细胞周期同步化。Westernblot做蛋白表达水平的检测。结果:50μmol/L的OAG使3T3细胞周期阻断在G2/M期。OAG对p34cdc2的表达没有影响,但可以使cyclinB的表达减少,对照组与处理组差异显著。结论:PKC激活剂OAG通过降低cyclinB的表达引起p34cdc2的活性改变而调节细胞周期。
Objective: To investigate the effects of protein kinase C(PKC) on the key regulation factor maturation promoting factor (MPF) in cell cycle procession of NIH3T3 cells. Methods: NIH3T3 cells were synchronized by aphidicolin. Flow cytometry was used to detect the process of cell cycle. Western blot was used to determine the change of p34^(cdc2) and cyclin B in level of translation on the G2/M bounder. Results: 1-oleoyl-2-acetylglycerol (OAG) of 50μmol/L didnt change p34^(cdc2), but deduced the cyclin B in level of protein expression. Conclusion: OAG can modulate the activity of p34^(cdc2) by deducing the expression of cyclin B.
