摘要
目的:探讨大鼠脑出血(ICH)后基质金属蛋白酶9(MMP9)的表达和脑含水量变化的关系。方法:采用未抗凝新鲜自体股动脉血注入大鼠尾状核建立脑出血动物模型。湿重干重法测定脑含水量的变化。结果:MMP9在ICH后6h开始表达,24~48h最明显(P<0.01),6d仍有较低水平表达。脑组织含水量ICH后6h开始增加,24~72h达高峰(P<0.01),6d基本恢复正常水平。结论:MMP9可能参与急性期脑水肿的形成。
Objective: To investigate the relationship between the expression of matrix metalloproteinase-9 (MMP-9) and the changes of cerebral water content in rats after intracerebral hemorrhage (ICH). Methods: Animal model of ICH was made through stereotactical infusion of 75 μl autologous fresh blood. Expression of MMP-9 protein was detected by zymogram analysis after 6 hours to 10 days. The cerebral water content was measured by using dry-wet weight method. Results: The expression of MMP-9 and cerebral water content in the hemorrhage site increased progressively after ICH. After 6 hours, significant (P<0.05) cerebral water content and MMP-9 activity were observed and reached the maximum level 24 to 72 hours (P<0.01). Cerebral water content returned to basal level at the 6th day. Conclusion: The MMP-9 may play a role in the pathogenesis of brain edema.
