摘要
用DNA-mRNA原位杂交和常规细胞生物学方法研究重组人转化生长因子-β_1(rh-TGF-β_1)对J6-1,J6-2和Namalva细胞的作用。rh-TGF-β_1对J6-1细胞增殖抑制作用呈明显的剂量依赖和双相时间依赖关系。对J6-1细胞的抑制主要通过抑制DNA合成,对J6-2的抑制则通过抑制DNA合成和促进凋落,Namalva细胞对它不敏感。rh-TGFβ_1对细胞基因表达的影响也因株而异:同时下调三个细胞系c-myc、LIF和p53的表达,上调v-sis的表达;对J6-2和Namalva的众多基因表达以下调为主,却上调J6-1细胞多个癌基因的表达,提示TGF-β可能使某些白血病细胞更趋恶性。
NA-mRNA in situ hybridization was used to demon-strate the effects of rh-TGF-β_1 on human leukemic celllines J6-1,J6-2 and Namalva. TGF-β_1 inhibited J6-1 cellproliferation in a dose-dependent and bi-directional time-dependent manner. Inhibition of cell division was foundin J6-1 cells , while both inhibition of division and pro-motion of apoptosis were observed in J6-2 cells whentreated with rh-TGF-β_1.No Significant change wasfound in Narnalva cells. The effects of TGF- β_1 on geneexpression varied among the cell lines.C-myc,LIF andp53 genes were down regulated while v-sis gene was up-regulated in all the 3 cell lines. TGF- β_1 down- regulatedmost gene expression in J6-2 and Namalva celis,but up-regulated most of them in J6-1 cells. It suggested thatTGF-β_1 might turn some leukemic cells more malignant.
