摘要
应用十二烷基硫酸钠－聚丙烯酰胺垂直板电泳及免疫印迹分析技术，以自制羊抗人载脂蛋白（ａ）［ａｐｏ（ａ）］为第一抗体，辣根过氧化物酶标记兔抗羊ＩｇＧ为第二抗体，建立了ａｐｏ（ａ）多态性检测方法，并对方法学中某些因素进行了探讨。检测了１９０名健康人血清，按Ｕｔｅｒｍａｎｎ等的分型方法，检出５种ａｐｏ（ａ）多态，共１２种表型。其中Ｂ占２．１％，Ｓ＿１占３．２％，Ｓ＿２占１８．４％，Ｓ＿３占２０．５％，Ｓ＿４占３０．５％。分子量范围在４８６０００～７４１０００之间，检出率为８３．２％。
olymorphic forms of human apo(a)were determined by SDS-PAGE electrophoresis and immunoblotting.According to the nomenclature of Utermann et al, the authors found 12 different phenotypes ranging in molecular weight from 486 000 to 74 1 000 in the healthy subjects. Among them B-type was 2.1%, S_1-type 3.2%,S_2-type 18.4%,S_3-type 20.5%,S_4-type 30.5%,“null”16.8%. The de-tective rate was 83.2%.
