摘要
采用CTAB法和SDS法提取烟草基因组DNA,经检测表明,CTAB法提取的基因组DNA纯度高于SDS法;CTAB法提取的DNA,其OD260/OD280值均高于1.8,相对分子量23kb左右,且能完全被EcoRⅠ和MseⅠ酶切消化,并能获得清晰的AFLP指纹图谱。
Two methods—CTAB method and SDS method were tested in isolation of tobacco genomic DNA. The results showed that the degree of purity of DNA extracted by using CTAB method was higher than that by using SDS method; the value of OD260/OD280 for the DNA samples obtained with CTAB method was over 1.8; the molecular weight of DNAs was more than 23kb, the DNAs were completely digestible with EcoR Ⅰ /MseⅠ, and the AFLP figure were obtained.
