摘要
目的研究环孢素A对小鼠脾细胞不同细胞因子生成的影响及其相互之间的关系。方法利用已建立的Th1细胞反应体系,ELISA法检测在不同浓度环孢素A条件下,BALB/c鼠脾细胞生成IFNγ、IL12及IL4的产量。结果环孢素A抑制IFNγ分泌的最佳浓度为10-1μg/ml,环孢素A抑制IFNγ分泌的同时不影响IL12的产量。高浓度(101μg/ml)环孢素A可促进鼠脾细胞分泌IFNγ和IL12。结论环孢素A对小鼠脾细胞分泌IFNγ有双向调节作用。高浓度环孢素A可促进鼠脾细胞分泌IL12。环孢素A对IFNγ的调节可能与IL12没有直接的关系。
Objective To study the influence of cyclosporin A on various cytokines production of mice spleen cells and their relationship each other.Methods Under various concentrations of cyclosporin A, the BALB/c mice spleen cells were cultured in in vitro model for Th1 type shifting. Then, IFN-γ、IL-12 and IL-4 produced by mice spleen cells were detected with ELISA.Results The optimal dose of cyclosporin inhibiting the IFN-γ production was 10-1μg/ml, but IL-12 was not influenced at the corresponding concentration of cyclosporin A. IFN-γ and IL-12 were increased by cyclosporin A at high concentration.Conclusion cyclosporin A modulated the production of IFN-γ of BALB/c mice spleen cells at double directions. High concentration of cyclosporin A could stimulated the production of IL-12. There was no direct relationship between IFN-γ and IL-12, When cyclosporin A regulated the immune reaction of mice spleen cells.
