摘要
目的:观察心肌梗死后心力衰竭雄兔体内雄激素水平变化特点。方法:实验于2003-09/2004-04在空军总医院动物研究中心和解放军总医院心内科实验室完成。实验选用60只4~5月龄健康雄性新西兰大白兔,随机将其分为模型组(n=52)和假手术组(n=8)。模型组进行冠状动脉结扎。造模3周后随机挑选10只模型兔继续观察到第15周,分别于造模前、心肌梗死后3,15周测定血清游离睾酮和硫酸脱氢表雄酮水平,并于同期行超声心动图检查评价左心室功能。结果:术后3周模型组、假手术组仍存活40,8只,15周时假手术组和模型组各存活7只。测兔血清雄激素水平时,由于部分兔子的血样不合格而剔除。①心肌梗死后(心肌梗死后3周即视为心力衰竭)3,15周模型组雄兔血清游离睾酮水平明显低于假手术组犤(100.02±23.99),(65.23±20.53)ng/L;(132.00±24.49),(127.50±19.00)ng/L,t=3.31,5.68,P<0.05犦。②心肌梗死后3,15周模型组雄兔血清硫酸脱氢表雄酮水平明显低于假手术组犤(0.060±0.014),(0.060±0.010)mg/L;(0.086±0.010),(0.087±0.020)mg/L,t=3.94,2.92,P<0.05犦。结论:从雄激素变化的角度讲,结扎兔冠状动脉后制作的心力衰竭模型具有复制人群心力衰竭的特征。本实验中心力衰竭雄兔血清游离睾酮和硫酸脱氢表雄酮水平显著下降,说明了雄激素水平下降是机体在心力衰竭发生发展过程中的内分泌反应之一。
AIM: To observe the changing characteristics of androgen levels in vivo in m ale rabbits with heart failure after myocardial infarction. METHODS:The experiment was carried out in the animal research center of Chine se PLA General Hospital of Air Force and the laboratory of Department of Cardiol ogy,General Hospital of Chinese PLA between September 2003 and April 2004.Sixty healthy male New Zealand rabbits of 4 to 5 months old were randomly assigned int o model group(n=52) and sham operated group(n =8).Ligation of coronary artery was performed in the model group. Three weeks after the establishment of models ,10 rabbit models were randomly selected for observation until the 15th weeks,an d the serum levels of free testosterone and dehydroepiandrosterone sulphate were determined before establishment of model and 3 and 15 weeks after myocardial in farction respectively,and echocardiography was undertaken to evaluate the left v entricular function at the same time. RESULTS:Three weeks after operation,40 and 8 rabbits were survived in the mod el group and sham operated group,respectively,and there were 7 survived rabbit s in each group after 15 weeks.The unqualified blood samples of some rabbits wer e excluded when the serum level of androgen was determined.① Three and 15 weeks after myocardial infarction (myocardial infarction for 3 weeks was considered a s heart failure),the serum levels of free testosterone in the model group were o bviously lower than those in the sham operated group[(100.02± 23.99),(65.23± 20.53) ng/L;(132.00± 24.49),(127.50± 19.00) ng/L,t=3.31,5.68,P< 0.05].② Thre e and 15 weeks after myocardial infarction,the serum levels of dehydroepiandrost erone sulphate were remarkably lower in the model group than in the sham opera ted group[(0.060± 0.014),(0.060± 0.010) mg/L;(0.086± 0.010),(0.087± 0.020) m g/L,t=3.94,2.92,P< 0.05]. CONCLUSION:Considering from the angle of androgen,a model of heart failure,wh ich is established after ligation of rabbit coronary artery,has the characterist ics of heart failure of duplicated crowd.In this experiment,the serum levels of free testosterone and dehydroepiandrosterone sulphate in the male rabbits with h eart failure are significantly decreased,which indicates that the decrease of an drogen level is one of the responses of endocrine during the occurrence and deve lopment of heart failure of organ.
