摘要
目的研究参麦注射液及其成分人参皂苷Rb1、Rg1对神经、血管内皮、Ⅰ型星形胶质细胞缺氧/缺糖再给氧损伤的保护作用,揭示参麦注射液脑保护作用的细胞机制,并初步确定其主要效应成分。方法培养神经、血管内皮、星形胶质三种细胞,含连二亚硫酸钠的无糖Earle’s液模拟造成缺氧/缺糖损伤,台盼蓝染色、MTT法、乳酸脱氢酶(LDH)漏出率评价药效。结果参麦注射液及其成分人参皂苷Rb1、Rg1对正常培养的三种细胞均无明显毒性损伤;与缺氧/缺糖再给氧损伤模型组比较,参麦注射液及人参皂苷Rb1、Rg1各组的LDH漏出率、细胞死亡率显著下降(P<0.001),细胞存活率显著提高(P<0.001~P<0.05)。结论参麦注射液的脑保护作用机制与其对神经、血管内皮、Ⅰ型星形胶质三种细胞缺氧/缺糖再给氧损伤的保护作用有关,人参皂苷Rb1、Rg1是其脑保护作用的主要效应成分;提示参麦注射液能够应用于中风病急性期的治疗。
Objective To study the protective effects of Shenmai Injection(SI) and its components,ginsenoside Rb_1 and Rg_1,on the damage of neurons,vascular endothelial cells(VEC) and astrocytes induced by the hypoxia/hypoglycemia and reoxygenation,reveal its mechanism of brain protective effect and to determine its main effective components. Methods To culture neurons,VEC and astrocytes and glycoprival Earle's fluid with sodium dithionate was used to establish the model of the damage induced by the hypoxia/hypoglycemia in vitro. The SI effect was evalusted by the methods of trypan blue stain,MTT assay and lactate dehydrogenase(LDH) leakage rate.Results SI and ginsenoside Rb_1 and Rg_1 have no obvious toxic effect on normal cultured three kinds of cells.Compared with the model group with the damage induced by the hypoxia/hypoglycemia and reoxygenation,the mortality rate and LDH leakage rate were decreased significantly(P<0.001),and the cell survival rate was increased significantly(P<0.001,P<0.05)in the groups of SI,ginsenoside Rb_1 and Rg_1. Conclusion The mechanism of brain protective effect of SI might be related to its protection against the damage of neurons,VEC and astrocytes induced by the hypoxia/hypoglycemia and reoxygenation. Ginsenoside Rb_1 and Rg_1 might be main effective components of SI.The result indicates that Shenmai Injection can be applied to the treatment of acute stroke in clinic.
