摘要 目的 探讨咖啡酸苯乙酯(caffeicacidphenethylester ,CAPE)对体外培养的大肠癌细胞HCT116细胞周期和cyclinD1表达的影响。方法 以人大肠癌细胞株HCT116为研究对象,PI染色、流式细胞仪检测CAPE处理2 4h后细胞周期分布,Westernblotting、RT PCR检测CAPE处理2 4、48h后cyclinD1表达的变化。结果 2 5、5 0、10mg LCAPE处理HCT116细胞2 4h后,G0 G1 期细胞百分率上升,S期细胞百分率下降,呈剂量依赖性。2 5、5 0、10mg LCAPE处理HCT116细胞2 4、48h后cyclinD1蛋白和mRNA表达均降低,呈剂量和时间依赖性。结论 CAPE诱导HCT116细胞周期阻滞,其机制可能与其下调cyclinD1表达有关。 Objective To study the effects of caffeic acid phenethyl ester (CAPE) on cell cycle, the expression of cyclin D1 in the cultured colorectal cancer cell line HCT116. Methods HCT116 cells were treated with CAPE at serial concentrations of 2.5, 5 and 10 mg/L. Cell cycle was analyzed by using flow cytometry (FCM) with propidium iodide (PI) labeling method. The expression of cyclin D1 was detected by Western blotting and RT-PCR. Results FCM analysis showed that G_0/G_1 phase rate increased, S phase rate decreased after HCT116 cells were exposed to CAPE (2.5, 5 and 10 mg/L) for 24 h, which were in a concentration-dependent manner. The mRNA and protein expressions of cyclin D1 decreased after HCT116 cells were exposed to CAPE for 24, 48 h, which were in a concentration- and time-dependent manner. Conclusion CAPE induced cell cycle arrest, which mechanism may be related to the significant down-regulation of cyclin D1 gene and protein expressions.
