摘要
目的:研究胃癌组织与癌旁组织中的环氧化酶-2(COX-2)的表达,COX-2抑制剂Celecoxib对胃癌发展的影响。方法:采用SP免疫组织化学染色检测胃癌组织与癌旁组织中的COX-2表达;细胞计数法检测Celecoxib对胃癌细胞SGC7901和BGC823生长曲线的影响;MTT比色法检测Celecoxib对SGC7901细胞和BGC-823存活率的影响;ELISA法测定Celecoxib对胃癌细胞SGC7901细胞和BGC823分泌释放VEGF蛋白的影响。结果:胃癌组织与癌旁组织中的COX-2表达存在显著差异。Celecoxib对胃癌细胞SGC7901、BGC823均有抑制作用,表现出剂量依赖性和时间依赖性。Celecoxib对胃癌细胞SGC7901、BGC823分泌VEGF均有抑制作用,随着药物浓度的增加和作用时间的延长,其VEGF分泌量也相应降低。结论:胃癌组织中的COX-2表达高于癌旁组织,Celecoxib可以抑制胃癌组织的发生发展。
Objective:To investigate the expressions of COX-2in gastric carcinoma and carcinoma side tissue,the relationship of cycloxyˉgenase-2inhibitor Celecoxib to gastric carcinoma.Methoeds:Detect the expression of COX-2in gastric carcinoma and carcinoma side tissue through SP immunohistochemical method.Apply cell counting to detect the effect of Celecoxib to the growth curve of gastric carcinoma cell SGC7901and BGC-823.Apply MTT chromatometry to detect the influence of Celecoxib on the survival rate of the cells SGC7901and BGC-823.Apply ELISA to detect the influence of Celecoxib on exudation of VEGFprotein of gastric carcinoma cell SGC7901and BGC823.Results:The expression of COX-2is markedly different betwent gastric carcinoma and carcinoma side tissue.Celecoxib has inhibitor effect to gastric carcinoma cell SGC7901?BGC823,and presents dose dependence and time dependence.Celecoxib has inhibitor effect to exudation of VEGF protein of gastric carcinoma cell SGC7901and BGC823.With the increasement of drug concentration and the prolonging of effect time,the exudation amount of VEGF also reduces correspondingly.Conclusion:The expression of COX-2in gastric carcinoma tissue is higher than in carcinoma side tissue,and celecoxib can restrain the occurrence and development of gastric carcinoma tissue.
