抗纤复方I号对酒精性肾损害PDGF-B表达的影响

The effect of kangxian fufang I on the expression of PDGF-B in alcoholic kidney damage

目的:探讨抗纤复方I号对酒精性肾损害时血小板源性生长因子-B(platelet-derivedgrowthfactor-B,PDGF-B)表达的影响。方法:按本科室创建的方法用乙醇及抗纤复方I号直接灌胃,于4周、8周、12周末处死大鼠,应用HE染色、透射电镜等观察其病理改变;用免疫组化方法检测中药组、酒精组、正常组肾组织PDGF-B的表达。结果:中药组同酒精组相比较,随着抗纤复方I号干预时间的延长,HE染色可见肾小球系膜增殖、间质局灶性炎性细胞浸润、肾小管上皮扁平化和小管扩张等病理改变出现延迟且明显减轻。免疫组化染色,中药组PDGF-B表达也明显低于酒精组(P<0.05)。结论:抗纤复方I号有效地防治酒精性肾损害,可能是促进了酒精的代谢,或干预了致肾间质纤维化的细胞因子PDGF的生成。

Objective:To explore the effect of kangxian fufang I on the expression of PDGF-B in experimental alcoholic kidney damage.Methods:By means of continuous intragastric ethanol and kangxian fufang I administration,we established the animal model.The animals were killed at the end of4、8、12weeks respectively.We observed the pathological changes by HE staining and transmission electron microscope.The expression of PDGF-B in herbal group,alcohol group and normal group was respectively measured by immunohistochemical method.Results:Hyperplasia of mesangium,local interstitial infiltration of inflammatory cell,the degeneration of renal tubular epithelial cell,the dilatation of tubule and other changes could be observed in alcohol group.Compared with it,in herbal group these pathological changes appeared late and were atˉtenuated as time progressed.Simultaneously the expression of PDGF-B in herbal group was significantly lower than that in alcohol group(P<0.05).Conclusion:Kangxian fufang I can effectively prevent and cure alcoholic kidney damage.It can possibly accelerate the matabolism of ethanol or intervene in the production of PDGF which can induce tubulo-interstitial fibrosis.
