摘要
血小板生长因子(PDGF)是诱导胰星状细胞(PSCs)活化的重要细胞因子,其分子机制尚不清楚。用PDGF-BB诱导体外培养的PSCs,旨在探讨PDGF信号转导通路在胰纤维化形成的分子发生机制中的可能作用。PSCs与不同剂量PDGF-BB(10ng/mL,25ng/mL和50ng/mL)共培养,24h后收集细胞,采用Westernblotting检测PSC中磷酸化细胞外信号调节激酶-1(pERK1)蛋白水平和RT-PCR检测Colα1(I)mRNA的表达。结果表明,不同剂量PDGF-BB作用于PSCs后,各组pERK1蛋白表达比空白对照组有不同程度上调;Colα1(I)mRNA在PSCs中表达也明显上调,且在一定范围内与PDGF-BB剂量呈正相关。
The mechanism of pancreatic stellate cells (PSCs) activation induced by platelet-derived growth factor (PDGF) is largely unknown. The purpose of this study is to investigate the influence of PDGF mediated signal transduction pathway on pancreatic fibrogenesis. Cultured PSCs were treated with 10 ng/mL,25 ng/mL and 50 ng/mL PDGF-BB respectively. PSCs were collected after 24 h co-incubation;the protein of phospho-extracelluar signal regulated kinase-1(pERK1) was detected by Western blotting;and Colα1(I)mRNA was detected by reverse transcription-polymerase chain reaction(RT-PCR). It is found that the expressions of pERK1 protein and Colα1(I)mRNA increase in PSCs induced by PDGF-BB, and a potential positive correlation between gene expression and administration dose of PDGF-BB is confirmed.
