EFFECTS OF Rb GENE ON INHIBITION OF CANCER GROWTH BY GENISTEIN IN HUMAN BREAST CARCINOMA CELLS

摘要 目的:研究Rb基因在染料木黄酮(genistein,Gen)抑制人乳腺癌细胞MCF-7细胞增殖中的作用。方法:采用MTT实验和集落形成实验观察Gen对人乳腺癌细胞增殖影响,Westernblotting检测Rb基因、细胞周期素cyclinE蛋白表达情况,RT-PCR检测Rb基因mRNA的表达。结果:Gen显著抑制MCF-7细胞生长,促进细胞Rb基因在蛋白及mRNA水平的表达,抑制cyclinE蛋白的表达。结论:Gen抑制人乳腺癌细胞增殖,其机制可能通过上调Rb基因表达,下调cyclinE蛋白表达。 Objective: To investigate the role of Rb gene in inhibiting breast cancer MCF-7 cells induced by genistein. Methods: In vitro experiments, MTT assay and clonal formation experiment were used to determine the cell growth inhibitory effect. Western blotting was performed to detect the protein expression of gene Rb and cyclin E. RT-PCR was performed to detect the mRNA expression of gene Rb. Results: Genistein inhibited the growth of breast carcinoma cell line MCF-7 in dose- and time-dependent manners. Genistein could improve the expression of Rb gene and reduce the expression of cyclm E gene. Conclusion: Genisein is able to inhibit the growth of breast cancer cells by up-expression of Rb gene and down-expression of cyclin E gene.
