毫米波辐射后小鼠肝H_(22)腹水型癌细胞AgNOR的形态定量研究

摘要 以毫米波治疗仪照射腹腔已接种H22肝癌细胞小鼠腹部,20min/d,连续15d。发现毫米波辐射组(GⅡ,6.90±0.28)、环磷酰胺组(GⅢ,6.54±0.69)和毫米波照射加环磷酰胺组(GⅣ,5.73±0.22)小鼠的癌细胞核仁组织区均有不同程度的嗜银蛋白颗粒减少和癌细胞坏死,其数量明显低于对照组(GⅠ,9.85±0.74)。Feulgen染色的标本经LeitzMPV-3显微分光光度计测定分析DNA,也发现6Ⅱ组(58.71±3.12)、GⅢ组(52.35±5.93)和GⅣ组(34.85±1.17)的癌细胞DNA显著低于GⅠ组(73.87±4.96)。结果表明,毫米波辐射可能降低H22腹水型癌细胞rDNA转录活动和蛋白合成。 The present study was performed by transplanting hepatoma H-22 ascitic cancer cells into mouse abdomen,then irradiated by millimeter wave(36.4GH2z, 105 mw, 8 mmwave length ,consecutive wave) 20 minutes each day for 15 days. Animals were dividedinto four groups : group 1 control(GⅠ),group 2 millimeter wave irradiation(GⅡ)group Ⅲ cyclophosphamide (GⅢ),and group Ⅳ millimeter wave irradiation plus cy-clophosphamide (G Ⅳ ).It was found that there were some degenerative changes of cancercells and a varied decrease in the mean number of AgNOR granules in the cancer cells of 6Ⅱ(6. 90±0. 28),6Ⅲ(6. 54±0. 69), and G Ⅳ(5. 73±0. 22) compared with G Ⅰ(9. 85±0. 74). There were significant differences between the three groups and G Ⅰ.The DNA content of Feulgen-stained hepatoma H- 22 ascitic cancer cells of the mice in the above four groups was also analyzed at the same time by Leitz MPV-3microspec-trophotometer, It was also found that there was a significant decrease in the DNA content ofG Ⅱ 57. 71±3.12),GⅢ(52. 35±5. 93),and G Ⅳ (34. 85±1. 17) compared with 6Ⅰ(73. 87± 4. 96). The results showed that millimeter wave irradiation could decrease he rDNA transcriptive activity and protein synthesis in the hepatoma H-22 ascitic cancer cells.
