摘要
目的:探讨针刺建立兔膝关节创伤性滑膜炎动物模型的方法及对该模型的评价。方法:将48只新西兰大白兔随机分为4组,每组12只。麻醉,针灸针垂直刺入兔后肢股四头肌中点和腓肠肌中点,连接电针仪,频率50Hz,强度1.5kPa,使其膝关节不停地被动屈伸,每天2次,共10天。每天测量兔造模肢膝关节周径的变化、针刺后跛行时间、关节表面温度变化等指标。造模实验结束后,ECT扫描检查判断关节局部的炎症情况;用1ml生理盐水注射入兔膝关节腔,回抽关节液,检测炎症因子IL-1β、PGE2、SOD、MDA的水平;取出滑膜,观察滑膜组织的病理切片、免疫组化等指标。结果:造模肢的各项检测指标和正常肢比差异有显著性意义(P<0.05,P<0.01),滑膜组织病理切片结果呈明显的炎性渗出性改变且随着针刺时间的增加改变更为明显。结论:该方法造模成功,模型经济实用。
Objective:To discuss and evaluate the method of establishing the model of synovitis on rabbit′s knee joint. Method:48 rabbits were devided into 4 groups at random, each group had 12. Rabbits were anaesthetised with 3% sodium pentobarbital. The rabbit′s hindlimbs were sticked vertically at the midpoint of quadriceps femoris and gastrocnemius muscles, connecting electroacupuncture apparatus, 50Hz with a frequency, 1.5mA intensity. Causing their knee joints did passive flexion and extension without intermission, twice a day,ten days. Measuring the change of rabbit′s knee joint circumference, the time of limp after being stimulated, the change of joint surface′s temperature etc. Estimating the inflammation of knee joint locally with the test of ECT,injecting 1ml Saline into the knee joint cavity,draw out the joint fluid to test the level of inflammation factors such as IL-1β,PGE2,SOD,MDA. Synovium was removed for pathological section,immunohistochemistry etc. Result:Every detecting markers of model limbs were notable significance compared with the normal limbs(P<0.05,P<0.01),the pathological section of synovium showed inflammational bleeding. Moreover,the longer the time of sticking, the more significant. Conclusion:The model of synovitis of rabbit's knee is successful. The method is feasible, economical applicable, and worth popularizing.
