摘要
目的研究丙烯腈(ACN)对大鼠胚胎中脑神经细胞增殖分化的影响。方法16d的大鼠胚胎中脑组织经取材、分离、消化后作原代细胞培养,加入不同浓度(0.01,0.1,1.0,10.0,50.0,100.0,200.0μg/ml)的ACN,从细胞形态学及细胞计数角度观察细胞生长与分化过程,同时测定蛋白质含量、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性并与对照组进行比较。结果不同浓度的ACN明显抑制胚胎中脑神经细胞的增殖和分化,集落形成率明显减少,细胞体积小;其半数分化抑制剂量(ICD50)为29μg/ml,半数存活抑制剂量(ICV50)为42μg/ml,均显示剂量效应关系。结论ACN能抑制胚胎中脑神经细胞增殖和分化,可能与其能抑制蛋白质合成,并引起脂质过氧化有关。
Objective To study the effects of acrylonitrile(ACN)on proliferation and differentiation of embryo midbrain nerve cells in rats.Method Primary culture of embryo midbrain nerve cells from rats at 14 days old was performed after being separated and digested.ACN at doses of(0.01,0.1,1.0,10.0,50.0,100.0 and 200.0)μg/ml was added to the culture,respectively.Proliferation and differentiation of the cultured cells were observed with cytomorphological method and cell counting.Contents of protein and malondialdehyde(MDA)and activity of superoxide dismutase(SOD)were determined for the culture and compared with the control.Result Proliferation and differentiation of embryo midbrain nerve cells in rats were obviously inhibited with ACN in a dose-dependent pattern,with significant reduction of cell colony formation and cell size.Median inhibitory concentration for differentiation(ICD_(50))was 29 μg/ml and median inhibitory concentration for viability(ICV_(50))was 42μg/ml.Conclusion Inhibition of acrylonitrile on embryo midbrain nerve cells in rats might be associated with its impact on protein synthesis and lipid peroxidation.
