摘要
F蛋白是近年来发现的肝细胞(?)志蛋白,它由肝细胞合成并存在胞浆内,是一种热不稳定、术溶性单链蛋白质,分子量44KD.等电点为6.5～6.7,在分子结构中至少有两个抗原决定簇.一个具种属特异性,另一个为人类特有。具有较强的抗原异性和免疫学活性。本文报道采用生化分离和柱层析技术从新鲜肝脏中提取纯化F蛋白,用凝胶电泳、免疫学和酶学方法进行鉴定等研完成果,为建立血清F蛋白放射免疫分析方法,和从体外刻度肝细胞损伤程度打下了基础。
A cytoplasmic liver-specific antigen now generally called F protein was first isolated by Fravial and Lindenmann from mouse liver extracts Recently, scientific researcher discover that F protein is a marker protein of liver cells. F protein is a 44 KDa protein is found mostly in the liver, PI 6. 5-6.7.unstable in heat, soluble mono-chain protein. There are over two antigenic determinants, one have species-specific characterization.another belong to human. They possess strongly antigenic and immune activaty. This paper reports the technique that isolation and purification of F protein from human liver. In order to set up radioimmunoassay of F protein and to study hepatocellular damage. Because serum F protein is a new sensitive and specific test of hepatocellular damage.
