CuSO_4对2.2.15细胞的影响 被引量:1

摘要 目的:探讨硫酸铜(CuSO4)对2.2.15细胞代谢及HBV复制和表达的影响及其机制. 方法:采用MTT法检测CuSO4对2.2.15细胞的毒性,流式细胞仪检测细胞周期.透射电镜法,Annexin V/PI双染后双变量流式细胞仪观察CuSO4对2.2.15细胞凋亡的影响,采用时间分辨荧光免疫分析技术(TRFIA)检测不同浓度的CuSO4作用后2.2.15细胞上清中HBsAg,HBeAg的P/N 值.Taq man荧光定量PCR检测不同浓度CuSO4作用后上清中HBVDNA滴度. 结果:MTT实验确定CuSO4在培养液中对2.2.15细胞的最大无毒剂量是128μmol/L.CuSO4作用后可引起细胞周期的改变,肝癌细胞被阻滞在G2+M期,且呈量效相关性.但是未发现CuSO4有诱导凋亡的作用,CuSO4干预12 d的结果显示2.2.15细胞上清中HBsAg,HBeAg的滴度及HBVDNA 的复制水平与对照组无明显差别. 结论:CuSO4有明显的细胞毒作用,可引起肿瘤细胞周期的改变及细胞坏死,提示了铜的抗癌作用的分子机制,CuSO4 短期处理2.2.15细胞后暂未发现其对HBV复制和表达的影响.
