存活素基因表达与膀胱癌细胞增殖活性的关系

Correlation between Survivin Gene Expression and the Proliferative Activity of Cancer Cells in Bladder Cancer

摘要 目的:探讨存活素基因在膀胱移行细胞癌中的表达,以及存活素基因表达与癌线胞的凋亡和增值活性的关系。方法:应用Western blot定量分析检测存活素蛋白在128例膀胱移行细胞瘤组织中的表达水平,原位DNA片段末端标记法检测肿瘤细胞的凋亡指数(Al),Ki-67单克隆抗体检测肿瘤细胞的增殖活性(Ki-67LI)。结果:Wstern blot分析结果表明在肿瘤组织中可检测到存活素的高表达,而正常膀胱组织中无存活素蛋白的表达。存活素蛋白的表达水平在肿瘤的临床分期和病理分级间差异具有显著性(P<0.05)。Spearman秩和相关检验提示存活素蛋白的表达水平与肿瘤的增值活性呈正相关(P<0.001),而与肿瘤细胞的AI无明显的相关性(P>0.005).结论:存活素蛋白表达在膀胱移行细胞癌的恶性进展中起重要作用,可作为评估膀胱移行细胞癌的恶性程度和预后的指标。 Objective: To identify the expression of surviving in bladder carcinogenesis and the correlation between surviving protein expression and the occurrence of spontaneous apoptosis, proliferative activity of cancer cells. Methods: Survivin protein level was quantified by western blot analysis in 128 cases of bladder transitional cell cancer (BTCC) tissue sample. The apoptosis index (AI) using in situ labeling apoptotic DNA fragment kit and the Ki-67 labeling index (Ki-67LI) with an anti-Ki-67 monoclonal antibody were analyzed in this tumors respectively. Results: High surviving levels were detected by western blot analysis in tumor tissue extracts. None expression of surviving protein was found in normal bladder tissues. Surviving levels were significantly different with different clinical stages and pathological grades of the tumors (P<0.05 respectively). Spearman rank correlation test revealed a positively correlation between surviving protein level and the proliferative activity (P<0.01) and feiled to find significant correlation between AI and surviving protein level (P>0.05). Conclusion: Survivin protein expression played an important role in the malignant progression of BTCC and could be sued as a marker of malignant potential and prognosis of BTCC.
