承气生血方对小鼠Lewis肺癌生长及转移的抑制作用

Inhibitory Action of Chengqishengxue Preparation on Growth and Metastasis of Lewis Lung Cancer of Mice

目的:观察承气生血方对小鼠Lewis肺癌肿瘤细胞生长及转移的抑制作用。方法:采用动物移植性肿瘤实验法,在C57BL小鼠腋皮下接种Lewis肺癌肿瘤细胞,口服给药后解剖动物,剥离肿瘤,计算肿瘤生长抑制率;取肺、肝、脾组织福尔马林固定,石蜡切片,HE染色后,显微镜观察肺转移结节,计算肿瘤转移抑制率,并观察肝脾形态学变化。结果:承气生血方1．2g·kg-1／d组与模型组比较肿瘤生长抑制率36．12％,p<0．01;1．2g·kg-1／d组及0．6g·kg-1／d组肿瘤转移抑制率分别为60％和62．5％,p<0．01。形态学观察,荷瘤小鼠肺转移瘤细胞核大,深染,形态不规则; 肝细胞肿胀,胞浆疏松,但未见肝组织有瘤细胞转移;脾细胞增大,排列不规则,可见大量多核巨细胞。结论:承气生血方具有抑制小鼠Lewis肺癌肿瘤细胞生长及转移作用。

Objective: To observe the inhibitory action of Chengqishengxue (carrying on qi and producing blood) preparation on the growth and metastasis of tumour cells of mice with Lewis lung cancer. Method: To implant the tumour cells of Lewis lung cancer under the axillary skin of C57BL mice by the experimentation of zoografting tumours, have an anatomy of the mice after orally taking Chenqishengxue preparation and cut the tumours off the lungs and calculate the inhibitory rate of the growth of the tumours; fix the tissues of the lungs, spleen and liver of the mice in formalin liquid and then observe the modules of tumour metastasis in the lungs under microscope through paraffin - section cutting and HE dyeing of the tissues, calculate the inhibitory rate of tumour metastasis and observe the morphocytological change of the mice' s liver and spleen. Result: Compared with the model group the inhibitory rate of tumour growth in the group of the mice fed by the dosage of 1. 2g·kg - 1/d of Chenqishengxue preparation is 36. 12% (P <0. 01) and the inhibitory rate of tumour metastasis in the two groups of the mice fed separately by the dosages of 1. 2g·kg - 1/d and 0. 6g·kg - 1/d results in 60% and 62% (P <0. 01) respectively. Through morphological observation under microscope the nuclei of metastatic tumour cells in the lungs of the mice are enlarged and thickly dyed and become irregular in shape; their liver cells appear swelling and their cytoplasms loose, but no metastasis of tumour cells is found in the liver tissues; and the cells of spleen are enlarged and irregularly arranged and a large number of multi - nucleated cells emerge among them. Conclusion: Chenqishengxue preparation has the functions of inhibiting the growth and metastasis of tumour cells of Lewis lung cancer in mice.
