调整下呼吸道谷胱甘肽水平与重构肺微循环治疗间质性肺病的实验与临床研究

The Experiment and the Clinical Research on Adjusting GSH Level of the Lower Respiratory Tract and Remodeling the Pulmonary Microcirculation

目的观察谷胱甘肽(GSH)与当归汤方剂合用治疗间质性肺病(ILD)的疗效。方法(1)动物实验,筛选100只健康SD大鼠,随机分为对照组、模型组、氢考组、GSH组、当归汤方剂组、合用组,除对照组外,其他各组气管内注入博莱平阳复制ILD模型,24h后各组分别给予相应药物,合用组每天给予等容积当归汤方剂10ml/kg灌胃及等容积GSH120mg/kg腹腔内注射,于第7、14、28d各处死5只大鼠,观察ILD病理变化及生化指标的改变。(2)临床应用,对71例ILD患者应用GSH与当归汤方剂联合治疗,观察临床症状、肺功能、影像学改变。结果实验动物与临床应用研究以合用组疗效为显著。结论GSH与当归汤方剂合用治疗ILD有明显疗效。

Objective To observe the curative effect of decoction angelicae sinensis and glutathione (GSH) on bleomycin-induced pulmonary fibrosis in rats and on patients with interstitial lung disease (ILD).Methods (1)Animal experiment:ninety-five SD rats in good conditions were divided into six groups randomly:control group,untreated group,hydrocortisone group,GSH group,decoction angelicae sinensis group and united group.The rats in all groups except the control intratracheally instilled with bleomycin (5 mg/kg body weight),and then treated with corresponding drugs respectively after 24 hours.The rats in the united group were treated with GSH(120 mg·~ -1kg·d~ -1,(intraperitoneally) and decoction angelicae sinensis (10 ml·kg~ -1·d~ -1,intrastomachly) until they were killed.On 7,14,28 days after instillation,five rats of each group were killed and the changes of lung histopathology and the level of GSH,hydroxyproline,hyaluronic acid in rat lung tissue and plasm were observed.(2)Clinical application:71 patients with ILD were treated with GSH and decoction angelicae sinensis,then the changes of their clinical symptom,pulmonary function and morphology were observed.Results Animal experiment and clinical application indicate that the therapeutic effect in the united group(GSH and decoction angelicae sinensis) was the most significant.Conclusion The curative effect of GSH united with decoction angelicae sinensis on ILD was the most significant.
