摘要
本文研究了食用仙人掌的离体培养所需的适宜培养基及其移栽技术,适宜的培养基为:不定芽诱导,2/3MS+BA0.5mg/L+NAA0.05mg/L,在诱导分化过程中,有些产生淡黄色颗粒状愈伤组织,从愈伤组织中分化出芽,有些直接分化出丛生芽;继代增殖,同诱导分化培养基相同,继代培养30d,平均增殖系数为4.0,分化率达100%;生根培养基为1/2MS+NAA0.2mg/L,培养一周后,开始生根,20d后,生根率达100%,平均每颗生根4颗。当根长达5cm时,移栽到石英砂,成活率达95%,再移栽到花盆,成活率也达96%。
The suitable medium in tissue culture of Opuntia milpa alta Haw and transplant techniques were studied in this paper. In the course of inducement and differentiation of the adventitious bud, 2/3 MS with the supplement of 0.5mg/L BA and 0.05mg/L NAA was the optimal medium. Some explants produced yellowish and granular calli. The buds was differentiated from the calli. Others don't produce calli, The buds was differentiated from the explant directly. The suitable medium for mulitiplication is identical with the inducement and differentiation. After 30 days, average mulitiplication coefficient is 4.0, differentiation percent is 100%.The buds began rooting after one week on 1/2MS with the supplement of 0.2mg/L NAA.After 20 days,rooting rate is 100%.When the length of roots reached 5cm, the seedling was transplanted into the quartz sand and then into the flowerpot. The survival rate of planting could be over 95% and 96% respectively.
