灰杨种群无性系范围及其相互关系的RAPD分析——以阿拉尔地区一个天然灰杨种群为例 被引量:8

Analysis on the Clone Extent and the RAPD of Its Correlation of a Natural Population of Populus Pruinosa——A Case Study in a Haturd Population of Populus Pruinosa in Aral

摘要 灰杨(Populuspruinosa)是荒漠河岸林的重要组成树种。应用RAPD技术对新疆阿拉尔地区的一个天然灰杨种群进行分析,通过10条引物对85个样品DNA扩增得到电泳图谱。结果显示:样品分属4个不同的无性系。所研究灰杨无性系的分布半径至少为350m,其分布范围约38hm2,证明灰杨具有逐步扩张、形成庞大种群范围的能力。 This paper discusses the population extent and the number of clones of a population of Populus pruinosa growing in the Aral region with the method of RAPD. The samples used in the study were sampled from 4 round plots with a delimited radius of 100 m in the Aral region at the upper reaches of the Tarim River, Xinjiang, and the trees of Populus pruinosa are 2~3 km away from the river channel. The electrophoretic profiles of 85 samples are analyzed with 10 selected random primes. The results show that the samples belong to 4 different clones, the growing radius of the clones of Populus pruinosa is 350 m at least, and the distribution area is about 38 hm 2. The results also show that Populus pruinosa has a capability to gradually expand and form huge population. The trees of clone A and clone B of Populus pruinosa in the study area grow concentratively, but their growing areas cover each other in the common boundary areas, and the spatial distance is shorter than that of the clone C and clone D; moreover, the genetic distance of clone A and clone B is short, and these 2 clones have closer affinity than the clone C and clone D. so it is conjectured that the clone A and clone B generate from the similar source. In the study area, the age of most trees of Populus pruinosa are over 10 years, however, there are no the 20~30-year-aged clones of Populus pruinosa, so it can be considered that the reproduction of Populus pruinosa has relied on root reproduction since recent 20~30 years at least. The sites of growing the trees of Populus pruinosa are lack in proper conditions for seed germinating, so that the trees of Populus pruinosa expand their population extent by cloning only, and the natural regeneration of the population of Populus pruinosa is extremely difficult after artificially destroyed. So it is important to reduce human disturbance so as to protect the growth and propagation of Populus pruinosa. The study results can provide the data for further researching the growth, propagation and ecological adaptability of Populus pruinosa.
