原子吸收光谱法测定丝裂霉素C-磁性纳米球中Fe_3O_4在小鼠体内的分布 被引量:2

摘要 目的:采用原子吸收光谱法测定丝裂霉素C磁性纳米球中Fe3O4在小鼠体内的分布。方法:用浓HNO3H2O2微波消化处理样品,乙炔空气火焰原子吸收法。结果:小鼠的心、肝、脾、肺、肾中低、中、高3种浓度加样回收率为98.80%~100.41%,日内精密度RSD均小于3%,日间精密度RSD均小于5%。经尾静脉给药后磁性纳米球主要被肝脏摄取;而在外加磁场的作用下磁性纳米球对肝脏的靶向率有明显的提高,至给药后60min是不施加磁场组的两倍。结论:提供了一种测定磁性靶向制剂中的磁性成分在体内分布的方法。 AIM:To establish atomic-absorption spectrometry methods for the Fe_3O_4 contained in mitomycin C magnetic nanoparticles in mice in vivo.METHODS:Samples were digested with 65% HNO_3-30% H_2O_2 and microwave and assayed by acetylene-air flame atomic-absorption spectrometry.RESULTS:Under the optimized conditions,the recoveries of heart,liver,spleen,lung,kidney were 98.80%~100.41%,the relative standard deviation RSD of intra-day of heart,liver,spleen,lung,kidney were all less than 3%,RSD of inter-day of heart,liver,spleen,lung,kidney were all less than 5%.Most of the magnetic nanoparticles were taken up by liver after the administration via tail vein.Under the guidance of an external magnetic field,targeting ratio of Fe_3O_4 to liver increased significantly.Targeting ratio of Fe_3O_4 to liver was two times larger than that of control group 60 minutes after administration.CONCLUSION:The method of determination of Fe_3O_4 in magnetic nanoparticles in mice in vivo was provided.
